    Comparative genomics and mutagenesis analyses of choline metabolism in the marine Roseobacter clade

    Choline is ubiquitous in marine eukaryotes and appears to be widely distributed in surface marine waters; however, its metabolism by marine bacteria is poorly understood. Here, using comparative genomics and molecular genetic approaches, we reveal that the capacity for choline catabolism is widespread in marine heterotrophs of the marine Roseobacter clade (MRC). Using the model bacterium Ruegeria pomeroyi, we confirm that the betA, betB and betC genes, encoding choline dehydrogenase, betaine aldehyde dehydrogenase and choline sulfatase, respectively, are involved in choline metabolism. The betT gene, encoding an organic solute transporter, was essential for the rapid uptake of choline but not glycine betaine (GBT). Growth of choline and GBT as a sole carbon source resulted in the re-mineralization of these nitrogen-rich compounds into ammonium. Oxidation of the methyl groups from choline requires formyltetrahydrofolate synthetase encoded by fhs in R.pomeroyi, deletion of which resulted in incomplete degradation of GBT. We demonstrate that this was due to an imbalance in the supply of reducing equivalents required for choline catabolism, which can be alleviated by the addition of formate. Together, our results demonstrate that choline metabolism is ubiquitous in the MRC and reveal the role of Fhs in methyl group oxidation in R.pomeroyi

    : Social life in state to International levels needs a system that governs how people should get along. The system setting makes people socially respectful and known as manners and etiquette. The implementation of ethical principle can also be applied in the implementation of public service including language, attitudes, and behavior of service providers. This study used qualitative research method. The location researched in this study was Dau District Office, Malang Regency focusing on the role of ethics in Public Service in Dau District Office, Malang, and how suitable the role of ethics and the development. To meet the objectives of this study, it used data analysis technique that involves three phases namely data reduction, data presentation, and conclusion. From the research results, it is showed that the role of ethics in public service in the Dau District Office of Malang is maximum; the officers are friendly, the services dos not that take a long time, and this office has also been awarded several times as the best service category. Furthermore, the role of ethics in accordance with the development has a good cooperation between the government and the society of Dau District, ranging from the early stages to the completion process, the community also participate in the implementation of the control and supervision of the construction by the existence of a good cooperation between the government and the society existing in Dau district, so that the development would change faster.     Limits on the HI content of the dwarf galaxy Hydra II

    Sensitive 21cm HI observations have been made with the Green Bank Telescope toward the newly-discovered Local Group dwarf galaxy Hydra II, which may lie within the leading arm of the Magellanic Stream. No neutral hydrogen was detected. Our 5-sigma limit of MHI < 210 solar masses for a 15 km/s linewidth gives a gas-to-luminosity ratio MHI/L_V < 2.6 x 10^{-2} Mo / Lo. The limits on HI mass and MHI/L_V are typical of dwarf galaxies found within a few hundred kpc of the Milky Way. Whatever the origin of Hydra II, its neutral gas properties are not unusual.Comment: Accepted for publication in Astronomy & Astrophysic

    Fluorescent artificial receptor-based membrane assay (FARMA) for spatiotemporally resolved monitoring of biomembrane permeability

    The spatiotemporally resolved monitoring of membrane translocation, e.g., of drugs or toxins, has been a long-standing goal. Herein, we introduce the fluorescent artificial receptor-based membrane assay (FARMA), a facile, label-free method. With FARMA, the permeation of more than hundred organic compounds (drugs, toxins, pesticides, neurotransmitters, peptides, etc.) through vesicular phospholipid bilayer membranes has been monitored in real time (”s-h time scale) and with high sensitivity (nM-”M concentration), affording permeability coefficients across an exceptionally large range from 10−9^{-9}–10−3^{-3} cm s−1^{-1}. From a fundamental point of view, FARMA constitutes a powerful tool to assess structure-permeability relationships and to test biophysical models for membrane passage. From an applied perspective, FARMA can be extended to high-throughput screening by adaption of the microplate reader format, to spatial monitoring of membrane permeation by microscopy imaging, and to the compartmentalized monitoring of enzymatic activity

    Face-Fusion of Icosahedral Boron Hydride Increases Affinity to γ‐Cyclodextrin: closo,closo‐[B₂₁H₁₈]⁻ as an Anion with Very Low Free Energy of Dehydration

    [Abstract] The supramolecular recognition of closo,closo‐[B₂₁H₁₈]⁻ by cyclodextrins (CDs) has been studied in aqueous solution by isothermal titration calorimetry and nuclear magnetic resonance spectroscopy. These solution studies follow up on previous mass‐spectrometric measurements and computations, which indicated the formation and stability of CD ⋅ B₂₁H₁₈⁻ complexes in the gas phase. The thermodynamic signature of solution‐phase binding is exceptional, the association constant for the γ‐CD complex with B₂₁H₁₈⁻ reaches 1.8×10⁶ M⁻Âč, which is on the same order of magnitude as the so far highest observed value for the complex between γ‐CD and a metallacarborane. The nature of the intermolecular interaction is also examined by quantum‐mechanical computational protocols. These suggest that the desolvation penalty, which is particularly low for the B₂₁H₁₈⁻ anion, is the decisive factor for its high binding strength. The results further suggest that the elliptical macropolyhedral boron hydride is another example of a CD binder, whose extraordinary binding affinity is driven by the chaotropic effect, which describes the intrinsic affinity of large polarizable and weakly solvated chaotropic anions to hydrophobic cavities and surfaces in aqueous solution.K.I.A. and W.M.N. are grateful to the DFG for grant NA-686/8 within the priority program SPP 1807 “Control of London Dispersion Interactions in Molecular Chemistry”.
